The Influence of macrophage co-culture to the fertility rate and blastocyst formation rate of the mouse's oocytes. Objective: 1. To setup a mouse IVF model. 2. To investigate the effect of co-culture with peritoneal macrophage to the fertility rate and blastocyst formation rate of the mouse's oocytes. Methods: 1. HMG 5IU, 10IU and 15IU were used in KunMing mouse aged 7-8 weeks, 10-14 weeks and over 15 weeks, weighted 32 gm, 32-40gm and over 40gm, respectively. HCG 10 IU was given 48 hours after HMG. Egg collection was performed 16-20hours later to evaluate the quantity and quality of the egg. 2. A total of 15 KunMing mouse aged 10-14 weeks were undergone ovulation induction and the oocytes were collected and randomly divided into three groups. The first group was the control group and cultured alone with no macrophage; the second group(experimental group 1) were co-cultured with macrophages right after the egg collection; the third group(experimental group 2) was cultured alone after egg retrieval, and co-cultured with macrophages from 8 cell stage. Results: 1. The biggest amount and the best quality of egg were found in Kunming mouse of 10-14 weeks of age and 32-40 gm of weight with HMG 10IU and HCG 10IU. 2. The fertility rate in experiment 1(72.0%) was significantly lower than that in control group(91.0%and 93.7%) in first group and experiment 2 respectively, (p<0.001). 3. Blastocyst formation rate of the fertilized oocytes in experimental group 2 (78.2%) was much higher than control group (50.3%, p<0.001). 4. Blastocyst formation rate of the oocytes in experimental group 2(73.2%) was much higher than control group(54.8%) and experimental group 1(50.0%); While the rate in experimental group 2 was the highest among the 3 groups, which was statistically significant. Conclusion: 1. Co-culture with peritoneal macrophages had an adverse influence on the fertilization process. 2. Peritoneal macrophages could enhance the mouse embryonic development and increase the blastocyst formation rate.
